Medvedev, a former world number one and 2023 Dubai champion, received a code violation for unsportsmanlike conduct and accused the umpire of bias against Russian players.
Griekspoor, ranked 47th, described the win as the best of his career and credited some luck in key moments, including during the second-set tiebreak.
The Dutch player will face fourth-seeded Stefanos Tsitsipas in the semifinals after Tsitsipas defeated Matteo Berrettini in three sets.
In the other semifinal, Felix Auger-Aliassime will take on Quentin Halys after victories over Marin Cilic and Luca Nardi, respectively.
