The 34-year-old previously announced his international retirement earlier this month, concluding a 95-cap career with England.
Marler has spent his entire 15-year professional career at Harlequins, making 285 appearances and winning two Premiership titles and a European Challenge Cup.
Known for his larger-than-life personality and on-field prowess, Marler has also been an advocate for mental health awareness during his career.
Harlequins head coach Danny Wilson praised Marler for his loyalty to the club and his significant contributions to both club and country.
