Adesanya had previously lost to Pereira twice in kickboxing and once in MMA, making this a revenge victory.
The main event capped off a major show in Miami that saw the retirement of UFC star Jorge Masvidal after a loss to Gilbert Burns.
Adesanya has dominated the middleweight division for the past four years and defended the title five times before losing it to Pereira.
The knockout cemented Adesanya as one of the greatest middleweights in UFC history.
